Analysis of limited-volume or limited- mass samples introduces an additional variable for the analytical chemist attempting to select the overall best method. While traditional LC has the precision, accuracy, and sensitivity characteristics that would be suitable for the vast majority of analyses, it requires injection volumes which might be several percent of the total sample volume available, especially in the case of experimental biologic compounds. Capillary LC, by comparison, requires a significantly smaller injection volume ( 20-200 nL), but total sample volume requirements are not straightforward due to injection complications which are encountered on the microscale. This presentation will attempt to define usable lower volume/mass limits for biologic samples that can be reliably used for analysis; first for a set of model set of phenones and then for peptide and protein mixtures.
