非酒精性脂肪肝(NAFLD)的发病率不断攀升。但是其发病机制尚不完全清楚,临床上更缺乏相应有效的防 治手段。而IRHOM2是一个有潜在的靶点,但其机制尚不完全清楚。在该研究中,确定了 USP13是IRHOM2的关键和新型内源性阻断剂,同时 USP13是IRHOM2相互作用蛋白,在肝细胞中催化IRHOM2的去泛素化。该项研究工作为NAFLD的治 疗以及USP13和iRhom2靶向药物的研发提供了崭新的思路。


V-15R具有10个转子可供选择,满足从细胞、细菌收集到蛋白、核酸提取的需求。本研究中使用了V-15R进行对消化和过滤后的肝组织原代细胞进行离心收集,方便后续实验步骤。


Cell culture and administration

Mouse primary cultured hepatocytes used in this experiment were isolated and concentrated from indicated experiments’ animals by liver perfusion. In brief, after anesthesia with sodium pentobarbital, mice abdominal cavity was opened. Then, the liver samples were tardily perfused with 1xliver perfusion working solution and 1xliver digest working solution  via the portal vein. Then, the digested liver tissue was filtered using a 100 mm steel mesh. The primary isolated hepatocytes were produced by harvesting the filter solution after 800 rpm centrifugation (Allegra V-15R, Beckman Coulter Life Sciences, Germany) (4 ℃, 5-10 min), and next purified using percoll-solution.

水资源短缺正成为一个全 球性问题。从传统灌溉系统到不充分灌溉系统的转变增加了土壤盐度,特别是粘土土壤。利用磁化水(MW)和生物刺激剂可以诱导植物对干旱和盐胁迫的抗性。本文评价MW和脯氨酸(P)对“太菲”石榴灌木生长、产量和果实品质的影响。总体而言,75% 作物需水量条件下的MW + P改善了盐渍粘土中石榴灌木的抗性机制,并提高了种植者的净利润。MW通常降低土壤盐分,表现为降低pH、EC、Na+和Ca2+的值。


利用V-15R高达20,000×g以上的离心力,对叶片研磨液进行离心澄清,方便后期对上清液中的脯氨酸进行定量。


Leaf Analysis

Sample (0.2 g) with 3 mL sulphosalicylic acid (3% w/v) using a porcelain mortar and pestle set, and then the mixture was centrifuged at 18,000× g for 15 min using a benchtop general purpose centrifuge Model Allegra V‐15R (Beckman Coulter Life Sciences, Indianapolis, IN, USA).

麸皮是小麦加工的主要副产品,开发利用率低。而多酚是麸皮中的主要生物活性成分,对人体健康有益。该文章对多酚提取条件和成分进行比较和研究,同时对多酚的抗氧化生物学性能进行了评价。


本研究中利用V-15R强劲的温控能力,对纤维素酶萃取处理的麦麸样品进行稳定的4 ℃离心,保证物质的生物活性。


Cellulose enzymatic extraction

Wheat bran was dissolved by adding ultrapure water at a material to liquid ratio of 1:21, adjusting the pH to 4.5 after adding 3% cellulase, extracting in a 50℃ water bath for 19 h, and then centrifuging at 10,000×g and 4 ℃ for 0.5 h (Allegra V-15 R, Beckman Coulter, Inc., USA). The supernatant was separated, and the filter residue was further subjected to ultrasonic-assisted extraction in 60% ethanol for 65 min at 800 W. At the end of centrifugation at 10,000×g and 4 ℃ for 0.5 h, the supernatant was separated. The two extracted supernatants were combined for further characterization.

纳米纤维素在干燥过程中结块导致其性能下降,是制约其生产和经济效益的主要因素。在本研究中,通过连续高盐-绿泥石工艺成功制备了二羧基纤维素纳米晶体(DCNC)。同时,将明胶和纳米纤维素经不同条件均质,冷冻干燥后用热水洗涤,获得了透光性和稳定性最 好的再分散DCNC悬浮液条件。本研究为纳米纤维素的再分散和储存提供了一条高效、便捷的途径,旨在实现其产业化。


V-15R的单次离心体积最 大可达3 L。本研究中使用了V-15R进行对氧化后的纤维素进行清洗,可以单次处理大体积的样品。


Sequential regioselective oxidations

Typically, 8 g of SFSC was soaked in water (250 ml) for 24 h at room temperature, next added 10 g NaIO4, 16 g NaCl. Meanwhile, the beaker was wrapped with several layers of aluminum foils to prevent entry of any light and the reaction was performed and stirred for 24 h. The oxidized fibers (dialdehyde cellulose, DAC) were thoroughly washed with distillated water by centrifugal (3000 g for 5 min, Allegra V-15R, Beckman Coulter International Trading (Shanghai) Co., Ltd.) until the conductivity of the supernatant was less than 30 lS/cm. The DACs were dried in a vacuum freeze-dryer.
