ThetestprincipleappliedinthiskitisSandwichenzymeimmunoassay.Themicrotiterplateprovidedinthiskithasbeenpre-coatedwithanantibodyspecifictoSecretoryImmunoglobulinA(sIgA).Standardsorsamplesarethenaddedtotheappropriatemicrotiterplatewellswithabiotin-conjugatedantibodyspecifictoSecretoryImmunoglobulinA(sIgA).Next,AvidinconjugatedtoHorseradishPeroxidase(HRP)isaddedtoeachmicroplatewellandincubated.AfterTMBsubstratesolutionisadded,onlythosewellsthatcontainSecretoryImmunoglobulinA(sIgA),biotin-conjugatedantibodyandenzyme-conjugatedAvidinwillexhibitachangeincolor.Theenzyme-substratereactionisterminatedbytheadditionofsulphuricacidsolutionandthecolorchangeismeasuredspectrophotometricallyatawavelengthof450nm±10nm.TheconcentrationofSecretoryImmunoglobulinA(sIgA)inthesamplesisthendeterminedbycomparingtheO.D.ofthesamplestothestandardcurve.
