Background:Vimentinisthemajorsubunitproteinoftheintermediatefilamentsofmesenchymalcells.Itisbelievedtobeinvolvedwiththeintracellulartransportofproteinsbetweenthenucleusandplasmamembrane.Vimentinhasbeenimplicatedtobeinvolvedintherateofsteroidsynthesisviaitsroleasastoragenetworkforsteroidogeniccholesterolcontaininglipiddroplets.VimentinphosphorylationbyaproteinkinasecausesthebreakdownofintermediatefilamentsandactivationofanATPandmyosinlightchain-dependentcontractileevent.Thisresultsincytoskeletalchangesthatfacilitatetheinteractionofthelipiddropletswithinmitochondria,andsubsequenttransportofcholesteroltotheorganellesleadingtoanincreaseinsteroidsynthesis.ImmunohistochemicalstainingforVimentinischaracteristicofsarcomas（ofneural,muscleandfibroblastorigin）comparedwithcarcinomaswhicharegenerallynegative.Melanomas,lymphomasandvasculartumorsmayallstainforVimentin.Vimentinantibodiesarethusofvalueinthedifferentialdiagnosisofundifferentiatedneoplasmsandmalignanttumors.Theyaregenerallyusedwithapanelofotherantibodiesincludingthoserecognizingcytokeratins,lymphoidmarkers,S100,desminandneurofilaments.
